The work will involve building a bridge on the Great Northern Highway to span the railway and road upgrades of the highway.
Clough and JV partner Acciona from Spain are currently working on the civil works contract awarded in May to build Koodaideri's northern rail formation.
As part of the work the JV will build a 170km-long rail spur to connect Koodaideri to Rio's existing rail line south of Lyre Siding.
Under the transport infrastructure contract the JV will build the rail bridge and 3.6km of road for the approaches to the bridge.
Side road upgrades will also be done at the Great Northern Highway and the Koodaideri mine access road intersection
Clough CEO and managing director Peter Bennett said the company was proud to continue being part of the ongoing development of WA's transport infrastructure after completing its first job with Main Roads in 1957.
